An air ambulance was called to the scene of the collision, which took place at 3pm today on the A143.
A spokesman for the ambulance service said the motorbike rider was in a serious but stable condition, while the woman who was riding pillion remains in a serious condition.
The two casualties were stabilised before they were taken to the James Paget University Hospital in Gorleston by ambulance.
The female driver of the car and the two passengers, who were children, were all unhurt.
Beccles Road in Fritton, which is near Great Yarmouth, remains closed by Norfolk police.
